Notwithstanding pode substituir in spite of porque ambos indicam contrariedade — isto é, expressam a ideia de apesar de.

Como uso, notwithstanding funciona como preposição quando vem antes de um substantivo ou expressão nominal:
Notwithstanding the rain, they went out. (A despeito da chuva, saíram.)

Também pode aparecer como advérbio em posição pós-positiva:
They were tired, their enthusiasm notwithstanding. (Estavam cansados, apesar do entusiasmo.)

Aplicando ao trecho do quadrinho:
